Supported Liquid Extraction (SLE)
Liquid-Liquid Extraction (LLE) is the process of distributing at two immiscible solvents. It plays an important role in sample preparation, however, as the speed of analysis going faster, sample preparation has become the bottleneck in bioanalysis. As the result, it is of importance to develop a simple, fast and efficient sample preparation technology in order to improve the quality of the sample analysis.
Supported Liquid Extraction (SLE) is a new sample preparation technique that is primarily used for the separation and purification of complex samples. We choose a porous material-calcined diatomaceous earth as solid support, which can replace the use of a separation funnel. Hydrometric with a high surface area and high capacity for aqueous adsorption makes an exceptional medium for SLE, which has the advantages of higher efficiency, simpler operation.
Comparing with traditional Liquid-liquid extraction technique, the features and benefits of SLE are as follows:
◇ Fast processing
◇ No emulsion formation
◇ Easy, safe clean-up with no
glassware and no separatory funnels
◇ No additional drying steps
after extraction
◇ Reduced technique dependence
◇ Higher reproducibility than
standard Liquid/Liquid Extraction
◇ Automation compatibility

Application Examples
Three compounds, methyl p-hydroxybenzoate, p-nitroaniline and benzoic acid, were used for extraction with SLE and LLE, respectively, and the extraction efficiency of the two was compared. The data are listed in the following table and figure. SLE is better than LLE in all cases.
