Filtration of proteins after their precipitation along with the capture of lipids is a routine sample preparation technique for the removal of proteins and lipids from plasma and serum samples prior to their analysis. This filtration-based protein precipitation (PP) method has gained popularity because it is simple and is an alternative to the traditional centrifugation-based technique using collection plates or micro tubes. However, when using PP as the sample preparation technique, the common membrane used in PP devices failed to block phospholipid, which is common in biological samples. The carry-over of phospholipid is one of the main problems in LC/MS-(MS) bioanalysis. Lipids that elute during the HPLC run can cause ion suppression and limit analyte sensitivity and detection in LC/MS. Over the course of multiple injections, lipids build up and bleed off the column, causing ion suppression, shifts in retention time and peak shape and necessitating additional column and system maintenance.  Thus, lipid and protein removal is especially important in a high throughput environment where short run times and ballistic gradients are insufficient to deal with these interferences chromatographically.
